If you are heading South, your next stop should be Montañita if you don't mind the smell of the "other type" of cigarets !
If you are not too attracted by this kind of ambiance, go straight to Guayaquil from where you might choose to go to Cuenca or to Loja and Vilcabamba.
And it's probable that even if you are heading North, going through Guayaquil is the best option anyway. From there you can go to Chimborazo or Baños.
Or if you want to go to Quito then do not go through Guayaquil, rather take a bus to Manta and then another one to Quito.
