From Quito as we already mentioned you can get anywhere you want.

We would advise you to go South first since only Otavalo is North and you might make some shopping there that you don't really want to carry all along your trip !!
So you should keep that for the last days, taking in account though that the market is on Saturday ! ! Is there a Saturday in your " last days " ?
So if you are going South ( which you would do anyway, even if you went before to Otavalo!), You can go to Cotopaxi or to Baños or maybe directly to Cuenca if you don't have too much time to spend in Ecuador.
