To get to Vilcabamba you have to get to Loja first.

You can get there from Quito or Guayaquil
Here are an approximation of the price you might pay.
The prices for adults in dollars include 12 % of IVA, the airport tax (2%) and the tax of security provided by the direction of divil aviation(1$)

The prices of the round trips are basically the doble of the prices of the one way trip.
Bus
The logical thing would be for you to get to Loja from Guayaquil or Cuenca or even coming from Peru.
Guayaquil-Loja:
Cooperativa de Transportes "Loja" will bring you there for 8 $ per person.
Cuenca - Loja:
Cooperativa de Transportes "Loja" must bring you there too. Count 6 hours.
Anyway there are buses every hour for 6 am to 10 pm. If it is not this company, it would be another.
Machala-Loja:
Cooperativa de Transportes "Nambija" will charge you 10 $ for the trip.
Quito-Loja:
Cooperativa de Transportes "Loja" will bring you there in....14 hours ( sorry for the bad news ).

Once in Loja, in the bus terminal go to the company ........ They will bring you to Vilcabamba for ....$ in a comfortable van which might be packed though.
